Need help identifying timing plate bolts251 CID Canadian T249 engine

Hi all. I was going to remove the plate behind the timing gears as its leaking behind it. No big deal but I ran into two items that I need help with in my picture. In yellow, What is this large screw for and does it need to be removed? Second the backwards bolt in Blue, do I really have to remove the old pan to undo this? The plate is still secure so something got to undo yet....am I missing something? Thanks Dale
